There are an immeasurable number of fungal species. Fortunately, only a handful cause harm to humans and the immune system is very adept at dealing with the fungal threat. However, in the immune compromised, such as those with underlying infections like COVID-19 or undergoing whole organ or stem cell transplants, fungal infections can be life-threatening. This issue has been further exacerbated by the emergence of antifungal drug resistance. Consequently, it is critical that we understand how fungal infections are recognized and cleared by the immune system to aid in the development of adjunctive immunotherapies. Here, we highlight the nonredundant role of innate immunity in controlling fungal infection with a particular focus on phagocytes and pattern recognition receptors, before describing the adaptive immune response comprising of dendritic cells, B and T cells. Finally, we overview the important advances in the development of immunotherapeutic strategies being employed for antifungal defense.
