Abstract
Unraveling the complex three-dimensional architecture within cells and tissues is fundamental to understanding the initial alterations which occur within disease processes. Localisation of the spatial arrangement of these organelles and their interactions at high resolutions can therefore provide information enabling early diagnosis and in turn, potentially lead to therapeutic breakthroughs. Transmission electron microscopy is a powerful tool to obtain ultrastructural detail at near-atomic resolution. For intermediate resolutions, there is great potential to use focused ion beams in combination with a scanning electron microscope to image cells and tissue at resolutions that are higher than what is possible with optical imaging methods. Here we provide an in-depth look at some of these methods and their practical application to problems in cell biology.
